Assessment of somaclonal variation in indirect morphogenesis-derived plants of Arracacia xanthorrhiza PAB

Abstract: The objective of this work was to induce and detect somaclonal variation in arracacha (Arracacia xanthorrhiza) plants regenerated via indirect morphogenesis, in order to evaluate the potential of this technique to produce new genotypes for breeding purposes of this crop. Calli were induced from petiole segments on Murashige & Skoog (MS) medium supplied with 0.1 mg L-1 2,4-dichlorophenoxyacetic acid. The regeneration of plants via indirect morphogenesis was carried out on half-strength MS medium without plant growth regulators. Fifteen randomly chosen plants were subjected to flow cytometry and “inter-simple sequence repeat” (ISSR) analysis. Background Yacon (Smallanthus sonchifolius) is a root crop native to the Andean region. Low sexual reproductive capacity is a major constraint facing the genetic breeding of this crop. Biotechnological techniques offer alternative ways to widen genetic variability. We investigated somaclonal variation in regenerants of yacon derived from in vitro somatic embryogenesis using simple sequence repeat (ISSR) analysis and flow cytometry. Results Twenty tested ISSR primers provided a total of 7848 bands in 60 in vitro regenerants and control plant. The number of bands for each primer varied from 3 to 10, and an average of 6.95 bands was obtained per ISSR primer. Chromosome number in Brazilian germplasm accessions of Paspalum hydrophilum, P. modestum and P. palustre (Gramineae; Paniceae) Genet. Mol. Biol.

This paper compiles results of chromosome counts of Paspalum hydrophilum, P. modestum and P. palustre. Four Brazilian accessions of P. modestum have shown 2n = 2x = 20 chromosomes, a number already found in one accession from Argentina and in two from Brazil. Three other Brazilian accessions showed tetraploid level (2n = 4x = 40), which was previously unknown in this species. In P. hydrophilum, only one of the accessions analyzed presented tetraploid level, initially established for the species from plants collected in Argentina. Five additional accessions from Brazil showed the diploid number, previously detected in a single Brazilian population. Cytogenetic analyses in Paspalum L. reveal new diploid species and accessions Ciência Rural

Chromosome numbers were counted in 126 new accessions of 50 Paspalum species from Brazil, Argentina, Paraguay and Bolivia. The chromosome numbers 2n=12, 20, 24, 30, 40, 50, 60, 80 were confirmed. Chromosome numbers for P. arenarium (2n=20), P. barretoi (2n=20), P. aff. ceresia (2n=40), P. corcovadense (2n=20), P. crispulum (2n=20), P. flaccidum (2n=40), P. nummularium (2n=20), P. scalare (2n=20), P. vescum (2n=20) and P. rectum (2n=20) and a diploid cytotype of P. malacophyllum are reported for the first time. Abstract The genus Capsicum comprises a large group of hot and sweet peppers, with different morphological varieties and degree of fruits pungency. Cytological and reproductive aspects in the Caespitosa group of Paspalum Ciência Rural

Somatic chromosome numbers are reported for thirty four germplasm accessions of Paspalum, Caespitosa group, representing five different species. All five species have shown x=10 as the basic chromosome number. The diploid 2n=20 chromosome number was confirmed for P. chacoense and P. indecorum, as well as sexuality for the latter. This is the first report of the chromosome number and cytological behavior for P. ligulare (2n=20 and 40), P. pleostachyum (2n=20, 30 and 40) and P. redondense (2n=20 and 40). The present results document regular meiosis in diploid accessions, with primarily bivalent pairing at diakinesis and metaphase I. ABSTRACT: In plant tissue culture system, the excessive supply of plant growth hormones may inhibit further growth and development of the regenerants and sometimes it may induce somaclonal variation. In this study, the optimum concentration of plant growth hormones for in vitro regeneration of Oryza sativa L. cv. MRQ 74 was identified and subsequently the occurrence of somaclonal variation of regenerated plantlets was determined through cytological analysis of in vivo and in vitro grown plantlets.
